that is probably just a pattern on his body. how old is he? if that spot is always the same shade when he is mad then it might be a problem but he should be fine if it changes shade when the rest of his body does.

It is most likely a pattern. Seeing as he is around 4-5 months old, he is probably getting ready to gain his adult colors. I wouldn't worry about it too much, but if it doesn't change color when he does, as turkenheimer said, then I would get professional help.

The spot doesn't changes with him and thats why i when to the vet already but even the vet doesn't know what it is ...but she is 100%sure that its not a tumor and it doesn't hurt him ..

Well I already did seek professional help.
And the vet doesn't know what it is.. She says it doesn't look like a bite mark neither.

Reasons why now I have posted my question in lots of different places looking for possibilities of what it could be and if anyone has seen it before ... and the vet doesn't think it's anything serious though just to keep an eye on it .. to make sure it doesn't grow or change places

It sounds like your husbandry is spot on although the basking temp might be a bit high. Here is what the panther care sheet says about temps:
Temperature:
Baby/juvenile (<9 months): ambient 72-80F (22-26C), basking 82F (28C)
Adult males: ambient 75-80F (23-26C), basking 85-90F (29-32C)
Adult females: ambient 75-80F (23-26C), basking 80-85F (26-29C)
It looks to me like there is a round reddish spot on his knee as well...or is it just the flash or pic?
I think that maybe, besides continuing to seek info from other panther keepers, the best you can do is keep an eye on your panther, the spot(s), and his behavior. He may be the only one that can give you the answers you are looking for.
Good luck, and please keep us posted.
